| I. Background | |
| CD80 基因（又稱 B7-1 或 CD28LG）是免疫系統(tǒng)中調控 T 細胞活化的關鍵分子，其編 碼的 CD80 蛋白屬于免疫球蛋白超家族，分子量約 60kD。該基因主要表達于活化的 B 細胞、 T 細胞、巨噬細胞及樹突狀細胞表面，通過與 T 細胞表面的 CD28 和 CTLA-4（CD152）結 合，提供雙向免疫調節(jié)信號：CD28-CD80 結合可激活 T 細胞增殖與細胞因子分泌，而 CTLA-4-CD80 相互作用則抑制 T 細胞過度活化，防止自身免疫反應。 CD80 基因的功能異常與多種疾病相關。腫瘤細胞常通過下調 CD80 表達逃避免疫監(jiān)視， 因此恢復 CD80 表達或使用 CD80 激動劑成為腫瘤免疫治療的新策略。同時，在自身免疫 疾病中，CD80-CTLA-4 通路的異?；罨赡軐е?T 細胞抑制過度，針對該通路的調節(jié)為疾 病治療提供了新靶點。 |
