| I. Background | |
| CD46 基因編碼的膜輔蛋白（MCP）是一種廣泛表達于所有有核細(xì)胞的跨膜糖蛋白， 屬于補體調(diào)節(jié)蛋白家族，在免疫穩(wěn)態(tài)和病原體防御中扮演關(guān)鍵角色。其核心功能包括：作 為補體系統(tǒng)的重要調(diào)節(jié)因子，通過輔助因子 I 裂解 C3b 和 C4b，保護宿主細(xì)胞免受補體過 度激活導(dǎo)致的損傷；同時作為 T 細(xì)胞共刺激分子，誘導(dǎo)調(diào)節(jié)性 T 細(xì)胞（Tregs）分泌抗炎因 子 IL-10，抑制過度免疫反應(yīng)，維持免疫平衡。 CD46 功能異常與多種疾病密切相關(guān)?；蛲蛔兛蓪?dǎo)致非典型溶血性尿毒癥綜合征 （aHUS），表現(xiàn)為微血管溶血性貧血、血小板減少和急性腎衰竭；在自身免疫性疾病中， 多發(fā)性硬化（MS）患者因 CD46 信號缺陷導(dǎo)致 IL-10 分泌不足，加劇神經(jīng)炎癥；類風(fēng)濕關(guān) 節(jié)炎患者則因 CD46 功能異常，T 細(xì)胞無法有效抑制促炎因子。此外，CD46 在腫瘤組織中 高表達，參與免疫逃逸機制，成為癌癥治療的新靶點。 |
